What is the dataset used in TMH Stab Pred?


A dataset comprising 38 α-helical membrane proteins has been curated from the MPTherm database, focusing on stability determinants. Firstly, the Unfolding Gibbs free energy change (ΔG0) derived from chemical denaturation data was considered, specifically in the absence of denaturant. Secondly, to ensure uniformity in physiological conditions, only data obtained within the experimental pH range of 5-9 and temperature range of 22-37°C were included. Lastly, to eliminate redundancy in protein measurements, the average (ΔG0) value was taken for instances where a protein had multiple measurements. This rigorous approach allowed for the creation of a robust and reliable dataset, setting the stage for accurate analysis and prediction of α-helical membrane protein stability.


What is the performance of TMH Stab Pred?


In our study, we evaluated the performance of our model through a jack-knife test, resulting in a remarkable correlation coefficient of 0.89 and a mean absolute error (MAE) of 1.21 kcal/mol. These metrics attest to the model's strong predictive capabilities for stability in α-helical membrane proteins. To validate the model's robustness and generalizability, we employed a blind test set. Encouragingly, the model demonstrated exceptional predictive accuracy on this test set, achieving an average MAE of 0.51 kcal/mol. This outcome underscores the model's reliability and potential practical utility in real-world scenarios, where accurately predicting α-helical membrane protein stability is of great importance.
